Besides being one of the best comics on the racks for both story and art, Darwyn Cooke's dialogue is the stuff of noir dreams:
Number Three: (Hussein Hussein to the Spirit)
"Mister Blue, you spear my heart with your pointy words."
Number Two: (Agent Silk Satin to a passed out Spirit)
"C'mon city boy, wake up. Don't make me give you a taste. Wake up."
Followed by one solid SMACK!

And finally, the Number One quote from The Spirit #4:(Silk Satin to the Spirit and Police Chief Dolan)
The Spirit: "Your men better be careful with Hussein. He's a slippery one."
Silk Satin: "Please. We're the CIA. Not some local cops and a Gainsborough."
Dolan: "Gainsborough?"
Silk Satin: "The blue boy. Google it."
